摘要
目的观察质粒PLXSN-bcl-2基因重组体对缺血性脑梗死大鼠的疗效及其对血管内皮生长因子(VEGF)的影响。方法将180只雄性W istar大鼠采用改良线栓法制成大脑中动脉阻塞(M CAO)2h模型,平均分为生理盐水组、空质粒组和bcl-2组,分别经侧脑室注入生理盐水、质粒PLXSN和质粒PLXSN-bcl-2,检测各组脑缺血再灌注3h、6h、24h、48h、72h后的脑梗死体积、bcl-2和VEGF蛋白表达状况。结果在同一时间点,(1)bcl-2组的脑梗死体积明显小于其它2组(P<0.01);(2)bcl-2组的bcl-2蛋白表达明显增多(P<0.001);(3)bcl-2组的VEGF蛋白表达明显增多(P<0.001)。结论bcl-2基因对VEGF蛋白的表达有明显促进作用,对缺血性脑梗死有明显治疗作用。
Objective To observe the therapy effect on the rats with ischemic cerebral infarction and the influence of VEGF by injecting the PLXSN-bcl-2 recombinant into their lateral ventricles. Methods The 180 healthy male Wistar rats were made into MCAO 2h models by reformed thread embolism method,and then we put them into the groups of saline,vacant PLXSN plasmid and bcl-2 meanly. Secondly, we injected saline.vacant PLXSN plasmid and PI.XSN-bcl-2 recombinant into the rats' lateral ventricle. Finally,we detected the cerebral infarction volume,the expression of bcl-2 and VEGF protein after 3,6,24,48,72 hours of cerebral ischemic reperfusion. Results (1)The infarction volume of the bcl 2 group was much smaller than the other two groups at the same time point(P〈0.01). (2)The expressed of bcl-2 protein in the bcl-2 group was far more than the other two groups at the same time point(P〈0. 001). (3)The expressed of VEGF protein in the bcl-2 group was far more than the other two groups at the same time point(P〈0. 001). Conclusion The method of injecting PI.XSN-bcl-2 into the rats' lateral ventricles has obvious therapy effect on ischemic cerebral infarction,and it promotes theex pression of VEGF protein greatly.
