米诺环素对糖尿病大鼠海马iNOS表达的干预 被引量:1

Effects of Minocycline on the expression of inducible nitric oxide synthase in the Hippocampus in a Diabetic Model of Rats

摘要 目的探讨诱导性一氧化氮合酶(inducible nitric oxide synthase,iNOS)在2型糖尿病大鼠海马中的表达情况以及米诺环素对iNOS表达的影响。方法高糖高脂饮食加小剂量链脲佐菌素建立2型糖尿病模型。将SD大鼠随机分为正常对照组、糖尿病模型组、米诺环素治疗组。造模成功1周后,米诺环素组每天腹腔注射米诺环素45mg/kg,糖尿病模型组和正常对照组腹腔注射等量的生理盐水,连续给药2周,然后取大鼠海马组织,免疫组化方法测各组大鼠海马中iNOS表达情况。结果免疫组化结果分析示正常对照组、糖尿病组、米诺环素组海马iNOS阳性神经元的平均光密度值(MOD值)分别为(0.101±0.012)、(0.196±0.020)和(0.128±0.010),与正常对照组比较,糖尿病组和米诺环素组iNOS表达的差异均差异有显著性(均P<0.01),糖尿病组和米诺环素组比较差异也有显著性(P<0.01)。结论iNOS在2型糖尿病大鼠海马神经元中表达异常增高,米诺环素可减少大鼠海马神经元iNOS的表达,这可能是米诺环素保护糖尿病认知功能障碍的机制之一。 【Objective】To explore the expression of inducible nitric oxide synthase (iNOS) in the Hippocampus in a type 2 diabetes model of rats and the changes after the treatment with Minocycline.【Methods】A model of type 2 diabetes mellitus was induced by a high-glucose and high-fat diet and low doses of STZ. Rats were then randomly divided into 3 groups:control group,DM group and Minocycline group. One week after the model was set up,Minocycline at a dose of 45/kg was administered intraperitoneally to the Minocycline group once daily for 2 weeks while the control group and the DM group were treated with the same dose of normal sodium. Then the Hippocampus were separated and detected the expression of iNOS with immunohistochemistry in each group.【Results】The analysis of the immunohistochemistry results were the MOD of the immunoreactive neuron in control group,MD group and Minocycline group were seperatedly (0.101±0.012),(0.196±0.020) and (0.128±0.010). Compared with the control group the expression was significantly increased in the DM group and the minocycline group (P〈0.01). While the expression was noticeably decreased in the Minocycline group in comparatson with the DM group (P〈0.01).【Conclusion】iNOS was significantly increased in the Hippocampal neuron of a type 2 diabetes model of rats. Minocycline could reduce the expression of iNOS in the Hippocampal neuron,which could be one of the mechanisms of Minocycline to protect the impaired cognition caused by diabetes.
