参芪固本方对小鼠Lewis肺癌的抑制作用 被引量:2

摘要 【目的】探讨参芪固本方对Lewis肺癌模型小鼠的治疗作用及机制。【方法】C57/BL系小鼠48只,随机分为参芪固本方组(剂量为0.5 g.只-1.d-1)、环磷酰胺组(剂量为80 mg.kg-1.d-1)、参芪固本方与环磷酰胺合用组(简称中西药合用组)及模型对照组;每只小鼠右腋窝皮下接种小鼠Lewis肺癌细胞混悬液(2×106/mL)0.2 mL,第2天开始参芪固本方组与中西药合用组以参芪固本方药液灌胃,连续10 d,第4天环磷酰胺组与中西药合用组腹腔注射环磷酰胺,连续3 d。末次给药24 h后称瘤体质量,计算抑瘤率;称胸腺和脾脏质量,检测外周血白细胞(WBC)数量、T淋巴细胞亚群及肿瘤坏死因子α(TNF-α)水平。【结果】参芪固本方可减轻荷瘤小鼠瘤体质量,抑瘤率达30.05%,与环磷酰胺合用后可使其抑瘤率由84.96%提升至90.02%;参芪固本方及中西药合用均可升高荷瘤小鼠T淋巴细胞亚群、TNF-α及外周血WBC水平(P<0.05或P<0.01),而参芪固本方单用可升高免疫器官脾脏和胸腺的质量。【结论】参芪固本方具有一定的抑瘤作用,可能与其增强荷瘤小鼠的免疫功能有关;与化疗药合用具有增效作用,并可拮抗其副作用。 pulmonary carcinoma in To investigate the therapeutic effect and mechanism of Shen Qi Guben Recipe (SQGR) on Lewis mice. [Methods] Forty-eight C57/BL mice were randomized into SQGR group (0.Sg/mouse/d), cytoxan (CTX) group (80mg/kg/d), SQGR + CTX group and model group. The mice were inoculated with 0.2mL (2 x 106/mL) of Lewis pulmonary carcinoma cell suspension by axillary subcutaneous injection. On 2^nd day of inoculation, mice in SQGR group and SQGR + CTX group were given gastric gavage of SQGR decoction and the treatment lasted 10 days; on the fourth day of inoculation, mice in CTX group and SQGR + CTX group were given intraperitoneal injection of CTX and the treatment lasted 3 days. Twenty-four hours after the last administration, the weight of tumor mass was detected to work out the tumor-inhibitory rate. Meanwhile, the weight of spleen and thymus gland, peripheral white blood cell (WBC) count, T lymphocyte subgroup and tumor necrosis factor a (TNF-a) level were examined. [Results] The tumor mass weight was reduced and the tumor-inhibitory rate was 30.05% in SQGR group. The tumorinhibitory rate was 84.96% in CTX group and 90.02% in SQGR + CTX group. Peripheral WBC count, T lymphocyte subgroup and TNF-α level were increased in SQGR group and SQGR + CTX group (P 〈 0.05 or P 〈 0.01 ) as compared with the model group. The weight of spleen and thymus has an inhibitory effect on Lewis pulmonary carcinoma function. SQGR has a synergistic action in combination gland was increased only in SQGR group. [ Conclusion] SQGR and its mechanism may be related to the increase of immune with CTX and can counteract the side effects of CTX.
