摘要
目的优化骨碎补总黄酮提取分离与纯化方法。方法以骨碎补总黄酮提取率及提取物中总黄酮含量为指标,优化骨碎补总黄酮提取工艺;以总黄酮的吸附与解吸特性为指标,筛选树脂。结果用10倍药材重量的体积分数为65%的乙醇提取3次,每次1.5 h为优化提取条件;非极性树脂D-101-1比较适合用于骨碎补总黄酮的纯化,其静态饱和吸附量与解吸率为305.3 mg.g-1和94.6%,动态饱和吸附量与解吸率为44.4 g.L-1和93.6%。结论骨碎补总黄酮提取物经D-101-1树脂纯化后,提取物中总黄酮的含量可达到66.0%,而上柱纯化前总黄酮的含量只有24.5%,说明采用D-101-1树脂纯化骨碎补总黄酮是可行的。
OBJECTIVE To optimize the extraction and the purification methods of the total flavonuid from Rhizoma Drvnariae. METHODS The extraction ratio and content of the total flavonoid in the extract were used as index to optimize the extraction process. The static/dynamic adsorption/desorption characteristics of total flavonoid were used as evaluation to select the better resin. RESULTS The optimum extraction conditions were, decided to be, 10 times of 65% alcohol to extract 3 times, 1,5 h once. D-101-1 macroporus resin gave good static/dynamic and adsorption/desorption characteristics of total flavnoid, which were 305.3 mg· g^-1, 44.4 g· L^- 1 and 94.6 %/93.6 %, so was selected to purify the total flavonuid from Rhizoma Drynariae. CONCLUSION Afler the purification proecss, the purity of the total flavonoid in the extract was up to 66.0%, while before purificatiun, it is just 24.5%. D-101-1 macroporus resin is suitable to purify the total flavonoid from Rhizoma Drynariae.
