摘要
目的研究富勒烯(C60)-甘氨酸衍生物在光照条件下对体外培养的小鼠骨肉瘤细胞生长的影响,探讨其作用机制。方法将不同浓度C60-甘氨酸与小鼠骨肉瘤细胞混合培养,用流式细胞仪检测C60-甘氨酸在光照条件下对小鼠骨肉瘤细胞的致坏死率。同时光照后检测产生的超氧阴离子自由基、超氧化物歧化酶(SOD)值的改变。结果随着C60-甘氨酸浓度增大,骨肉瘤细胞的坏死率增大,同时导致超氧阴离子自由基升高,SOD含量下降。结论C60-甘氨酸在光照条件下对小鼠骨肉瘤细胞有较强的杀伤作用,作用机制与自由基的产生有关。
Objective To investigate the effects of fullerenes(C60 )-glycin derivatives on the growth of mouse osteogenic sarcoma cells and the mechanism. Methods Mouse osteogenic sarcoma cell lines (Lm8) were co-cultivated with different concentrations of C60-glycin (0, 25, 50, 75 and 100 μg/ml) and then illuminated under the tungsten halogen lamp (500 W, 30 min, once a day, 3 days). Mortality of the cancer cells was inspected by flow cytomety (FCM). The contents of superoxide anion free radical and superoxide dismutase (SOD) were examined at the same time. Results Mortality of the cells was increased with the increase of C60-glycin's concentration, When the concentration of C60-glycin reached 100 μg/mL, (31.12±2.85) cancer cells died, Meanwhile, contents of superoxide anion free radicals were increased but SOD decreased with the increase of C60-glycin's concentration, Conclusion Under the condition of photoaction, mouse osteogenic sarcoma cell can be killed by C60- glycin, The mechanism is related to the production of free radicals,
