摘要
目的观察大豆异黄酮(SI)对大鼠心肌细胞动作电位的影响和培养心室肌细胞钙电流的作用。方法采用悬浮玻璃微电极法和全细胞膜片钳方法。结果SI能剂量依赖性地降低动作电位幅值(APA)、超射(OS)、最大除极速率(Vmax)、缩短复极50%及90%水平的动作电位时程(APD50、APD90);SI 0.1μg/m l和1.0μg/m l对L-型钙电流可见不同程度的抑制作用。结论SI可使心肌细胞动作电位除极参数降低,动作电位时程缩短,对钙电流的抑制作用可减轻心肌细胞钙超载,进而保护心肌。
Objective To observe the effects of soybean isoflavone (SI) on the action potential in rats cardiomyocytes and the L-type calcium current in cultured neonatal rat ventricular cells. Methods The glass suspension electrode and the whole cell voltage clamp technique were used. Results SI could reduce the action potential amplitude (APA), overshoot (OS), maximal velocity of phase 0 depelarizatian (Vmax) and repolarization action potantial duration (APD50 and APD90) in dose-dependence. SI (0.1, 1.0 μg/ml) had inhibition on the L-type calcium current. Condusions SI could reduce parameters of polarization of action potential and APD, alleviate calcium overload in myocardium by inhibiting calcium current to protect myocardium.
