摘要
以黄色短杆菌ATCC14067为出发菌株,经硫酸二乙酯,紫外线玫 硝基胍逐级诱变处理,α-氨基-β羟基戊酸,S-2-氨基乙基-L-半胱氨酸,磺胺胍,乙硫氨酸,α-氨基丁酸,异亮氨酸氧肟酸等氨基酸结构类似物及琥珀酸为碳源板定向筛选,获得一株L-异亮氨酸高产菌ZQ-4在含13.5%葡萄糖培养基中,摇瓶发酵72h,L-异亮氨酸积累可达2.8-3.0%。
Brevibacterium flavum ATCC14067 as a starting strain was stepwise treated with diethylsufate(DES), ultraviolet (UV) and N-methyl-N'-nitrosonguanidine (NTG). A L-isoleucine over-producing mutant,ZQ-4,was obtained, which could resist to α-amino-β-hydroxyvalic acid (AHV),S-(β-aminoethyl)-L-cysteine (AEC), sulfaguanidine (SG), ethionine (Eth), α-amino-butyric acid (α-AB),isoleucine-hydroxamate (IleHx) and could grow quickly on succinate (as sole carbon source) medium. ZQ-4 could accumulate 2.8-3.0% of L-isoleucine in a medium containing 13.5% glucose shaking at 31±1℃ for 72hrs'.
