加味宣肺透解剂对流感病毒感染小鼠肺组织中炎性细胞因子表达的影响 被引量:7

Effect of Jiawei Xuanfei Toujie Ji on the expression of Inflammation Cytokines in Lungs of Influenza Virus(FM1) Infected Mice

摘要 目的:探讨加味宣肺透解剂对实验性小鼠流感病毒肺炎的治疗及其药效机制。方法:复制流感病毒鼠肺适应株(FM1)小鼠肺炎模型,以加味宣肺透解剂灌胃治疗,共给药4天。感染后第5天,取各组小鼠肺组织福尔马林固定,石蜡包埋切片,HE染色,光学显微镜下观察肺组织病理变化;用SABC免疫组织化学法检测肺组织中TNF-α、IL-1β、MCP-1表达。结果:加味宣肺透解剂治疗组肺组织病变程度明显轻于病毒感染模型组(P<0.05),同时肺组织中TNF-α、IL-1β、MCP-1的表达明显低于病毒感染模型组(P<0.05)。结论:加味宣肺透解剂减轻肺部急性炎性损伤可能与该药降低肺组织中炎性因子TNF-α、IL-1β、MCP-1的表达有关。 Objective:To study the therapeutical effect of Jiawei Xuanfei Toujie Ji (JXT) on influenza viral pneumonia in mice and its mechanism. Methods:Mice were inoculated intranasaUy with mouse lung- adapted Influenza virus strain (FM1) ,and were treated with JXT. At 5th day after postinoculation,mice were sacrificed, and their lungs sections were stained with hematoxylin and ecsin. ( H&E), then pathologic changes of the mice lungs were observed under microscope. SABC immunohistochemistrial method was used to detect MCP - 1 ,TNF-α、IL-1β expression in lung tissue sections. Results: Compared with model group, the pathological changes of JXT group were ameliorated obviously(P 〈 0.05). Compared with model group, MCP - 1, TNF -α、IL-1β of JXT group were dramatically decreased( P 〈 0.05). Conclusion: The effect of decreasing the expression of MCP - 1, TNF -α、IL-1β in lung maybe one of the factors that JXT decreases the pathogenesis of influenza - induced acute lung injury.
