过度运动对肾脏细胞外基质、金属蛋白酶及其抑制因子影响的实验研究 被引量:6

Research on Effects of Excess Exercise on Renal ECM,MMPs and Inhibitors of MMPS

摘要 通过在不同运动负荷的运动训练状态下,观察大鼠肾脏组织CoI IV、Laminin蛋白、MMP-9酶的表达和尿TP排泄率的变化,以及不同的运动负荷对MMP-9、TIMP-1和ColIVmRNA基因表达的影响,研究不同负荷的运动训练对大鼠肾脏ECM、MMP-9/TIMP-1的影响以及MMP-9/TIMP-1的变化在导致肾脏组织结构、生理功能损害上的作用,进一步探讨运动性蛋白尿的产生机制。实验结果表明:经过8周的过度训练后,C组大鼠肾脏CoI IV、Laminin相对于A、B两组出现重度阳性表达,具有显著性差异:Col IV、MMP-9、TIMP-1mRNA表达显著增强,TIMP-1mRNA的表达上调了225%显著大于MMP-9mRNA表达上调147%。且尿TP排泄率也显著升高。说明过度运动训练导致ECM产生增加,降解能力下降,致使肾小球及肾小管基底膜ECM异常代谢、沉积和肾间质纤维化的发生和加剧,肾脏组织超微结构及生理功能受到损害。 In order to explore influences of different exercise intensity on renal ECM, MMP-9/TIMP-1 expressin, effects of changes of MMP-9/TIMP-1 on the harmful function to renal tissue structure and physiological function, therefore having a further study in the mechanism of the cause of exercise-induced urinary protein, the Col Ⅳ, LA,MMP-9 expression , the changes of urinary TP excretion rate, and MMP-9 , TIMP-1 and Col Ⅳm RNA expression were tested at different exercise intensity. The results of this study indicated that after 8 weeks over-load training, Col Ⅳ, MMP-9, LA expression, Col Ⅳ,MMP-9, TIMP-1m RNA expression and urinary TP excretion rate increased significantly in group C than in group A and B. TIMP-1m RNA expression in group C increases by 225%. It showed that over-load training induced the increase of ECM, decreased the degradation ability, and as a result it caused the abnormal metabolism, accumulation of ECM and damage to renal micro-structure and physiological function.
