摘要
目的探讨醒脑启智胶囊对小鼠脑组织海马区脑红蛋白(NGB)mRNA表达的影响。方法120只昆明种小鼠随机分为假手术组、模型组、醒脑液高剂量组、醒脑液低剂量组、银杏叶液对照组、尼莫地平液对照组,每组20只。采用双侧颈总动脉结扎反复脑缺血再灌注的方法制备血管性痴呆小鼠模型。术后7d进行行为学实验和脑组织的病理形态学观察,并采用反转录-聚合酶链反应(RT-PCR)检测实验小鼠海马组织NGBmRNA的表达。结果①行为学实验观察显示,模型组小鼠学习成绩与记忆成绩下降,各治疗组小鼠的学习成绩与记忆成绩均有提高(P<0.05或P<0.01)。②光镜下病理形态学显示,术后模型组小鼠脑组织海马区呈缺血性病理改变,各治疗组小鼠病变轻于模型组。③模型组小鼠脑组织海马区NGBmRNA的表达较假手术组增加(P<0.05);各用药组均较模型组增加(P<0.01)。结论醒脑启智胶囊对血管性痴呆小鼠有明显治疗作用,其作用机制之一是调节海马组织NGBmRNA的表达,减轻缺血再灌注损伤。
Objective To investigate the effects of Xingnaoqizhi capsule on the expression of NGB mRNA of hippocampus tissue in mice with experimental vascular dementia. Methods A total of 120 male Kunming mice were divided into 6 groups randomly: Sham operation group, model group, high dose Xingnaoqizhi group, low dose Xingnaoqizhi group, Extract of GinkgoBiloba (银杏叶) control group, Nimodipine control group. Each group included 20 mice. The models with vascular dementia were duplicated by the operation of cerebral ischemic reperfusion. At the 7th day after praxiology experiment, the mice were cut head to obtain cerebral tissue. The morphologic changes of cerebral tissue and the expression of NGB rnRNA of hippocampus tissue in mice were observed. Results ①Results of praxiology experiment showed that the learning and memorizing performance record in model group decreased and that increased in other group (P 〈 0.05 or P〈0.01). ② The pathomorphologic analyze indicated that hippocampus tissue of mice brain appeared changes of ischemia. The pathological changes in each treatment group was weaker than that in model group. ③ The expression of NGB mRNA of hippocampus tissue of mice in model group were higher than that in sham-owration group(P〈0.05), and that in each medical group was higher than that in model group(P〈0.01). Conclusion Xingnaoqizhi Capsule was effective on experimental mice with vascular dementia. The mechanisms might be that Xingnaoqizhi Capsule could regulate the expression of NGB mRNA of hippocampus tissue in mice.
