养心定悸膏对内毒素引起的大鼠急性肺损伤及休克的保护作用 被引量:1

摘要 【目的】观察养心定悸膏(DJG)对内毒素(ET)所致大鼠休克及肺损伤的保护作用。【方法】28只wistar大鼠随机分为ET组(n=11)、DJG组(n=11)和生理盐水(NS)组(n=6),ET组和NS组常规饲养,DJG组喂养含DJG饲料和水。饲养5周后,ET组和DJG组经静脉注射灭活大肠杆菌(2 000亿/kg),NS组经静脉注射生理盐水,观察注射后10、30、60、120 min大鼠存活率及3组大鼠血压、外周血白细胞总数及中性粒细胞百分比的变化情况。120 min处死大鼠,检测血浆丙二醛(MDA)浓度,观察肺组织病理形态改变,称量肺干/湿重比值,测肺组织匀浆中MDA、黄嘌呤氧化酶(XOD)、微量元素钨、钼含量。【结果】ET组大鼠存活率降低,其血压、外周血白细胞总数、中性粒细胞百分数、肺干/湿重比值较NS组均明显降低(P<0.05),肺组织严重水肿,肺内白细胞浸润增多。肺组织MDA、XOD含量显著增高(P<0.05)。DJG组大鼠与ET组比较,未出现大鼠死亡,其血压、外周血白细胞总数、中性粒细胞百分数以及肺干/湿重比值的下降幅度均明显降低,肺组织中钨/钼比值升高,肺组织MDA、XOD含量显著降低(P值均<0.05)。【结论】DJG对大肠杆菌内毒素所诱导的大鼠急性肺损伤和休克具有保护作用。 [Objective]To observe the protective effect of Dingjigao (DJG) on the acute lung injury and shock induced by endotoxin (ET) in rats. .[Methods]Twenty eight Wistar rats were randomly divided into three group : Endotoxin group( ET group, n = 11 ), DJG group ( n = 11 ) and normal saline group( NS group, n =t5). ET group and NS group were fed routinely. DJG group was given food containing Dingjigao. Five weeks later, ET group and DJG group were bolus injected (iv) inactivated E. coli (2× 10^11 organisms/kg), and NS group was bolus injected equivalent NS. The data of blood pressure, circulation white blood cells(WBC), percent of neutrophil were collected at 10, 30, 60, and 120 minutes after injection. MDA concentration of plasma and contents of MDA, XOD, wolfram and molybdenum in rat lung tissue were determined. The dry-wet weight ratio of lung, morphological changes, and survival rate were observed. [Results]In ET group, compared with NS group, total count of circulation WBC, percent of neutrophil, dry-wet weight ratio of lung reduced significantly(P〈0.05). Severe pulmonary edema and WBC infiltration were observed. The level of MDA and activity of XOD increased significantly( _P d0.05). Survival rate was low. In DJG group, all the changes observed above were attenuated(P 〈0. 05). [Conclusion]DJG could protect rats from the acute lung injury and shock induced by endotoxin.
