猪α1岩藻糖转移酶基因点突变的PCR-SSCP检测 被引量:2

Detection of point mutation of porcine α1-fucosyltransferase gene by PCR-SSCP approach

摘要 猪α1岩藻糖转移酶基因(FUT 1)位于猪第6号染色体上,是猪肠毒素大肠杆菌F 18受体(ETEC F 18R)理想的候选基因。采用PCR-SSCP法共检测外来猪种、国内地方猪种和培育猪种3个类型9个猪种共计255个个体FUT 1基因M 857位点的多态性,所检测的3个外来猪种均存在GG和AG两种基因型,而国内地方猪种中除临高猪外均表现为极端的单态分布,只存在GG基因型。采用PCR-SSCP方法对FUT 1基因点突变进行检测,具有试验成本低、操作简便和灵敏度高等优点,可望在育种实践中得到更广泛的应用。 αl-fucosyltransferase (FUT1) gene has been identified as a candidate gene for Enterotoxigenic Escherichia coli F18 receptor (ETEC F18R) and was located on the pig chromosome 6. By the PCR-SSCP method, the loci of 307 bp of FUT1 gene in 9 pig breeds for polymorphism were screened respectively. The results showed that the GG and AG genotypes were detected in three foreign breeds, however, the AG genotype could be detected only in Lingao pigs of all tested Chinese native pig breeds, and the others carried only genotype of M857^GG, and showed extremely abbreviation distribution. The method of PCR-SSCP was used in this research to detect the point mutation of FUT1 gene, this method has the advantage of low cost, easy manipulation, and high sensitivity. This method may be applied for commercial detection in swine breeding.
