靶向胰岛素样生长因子受体的RNA干扰技术抑制胶质瘤生长 被引量:1

摘要 目的研究靶向胰岛素样生长因子-1受体(IGF-1R)的小干扰RNA(siRNA)对胶质瘤生长的抑制作用。方法逆转录-聚合酶链反应和W estern b lot测定RNA干扰后IGF-1R表达水平;采用Transwell体外侵袭实验进行细胞体外的侵袭力分析;流式细胞仪检测细胞凋亡率;裸鼠成瘤实验测定IGF-1R基因表达下调后胶质瘤C6细胞在体内致瘤能力的改变。结果靶向IGF-1R基因的siRNA可以有效抑制IGF-1R基因的表达,使IGF-1R mRNA表达减少60%,蛋白表达减少50%;流式细胞术检测到细胞凋亡率明显升高(P<0.01);裸鼠体内成瘤能力降低。结论siRNA介导的IGF-1R基因下调能明显抑制胶质瘤细胞生长。 Objective To study the inhibitory effect of small interference RNA (siRNA) targeting insulin-like growth factor-1 receptor (IGF-1R) on the growth of glioma. Methods Transcripfion-polymerase chain reaction and Western blot were used to analyze the down-regulation of IGF-1R expression. The invasi'veness of C6 cell in vitro was measured by the matrigel invasion assays. Flow cytometry was performed to analyze cell apoptosis. The ability of the tumorigenicity in the nude mice was measured. Results The expression of IGF-1R was suppressed effectively by siRNA targeting IGF-1R. The expression of IGF-1R mRNA and IGF-1R protein decreased by 60% and 50% respectively. Cell apoptosis was induced from 1.29% to 15.88% (P 〈0. 01 ). The ability of tumorigenicity in the nude mice was reduced. Conclusion Down-regulation of IGF-1R by siRNA can suppress glioma cell growth markedly.
