摘要
目的检测组织工程化人工皮肤的细胞外基质成分——胶原蛋白的合成和含量的动态变化,为组织工程化人工皮肤的研究提供生物化学依据。方法利用壳多糖作为基质网架制备组织工程化人工皮肤,用3H-脯氨酸掺入法测定胶原蛋白合成,Woessner法测定胶原蛋白含量。结果随着培养时间的延长,组织工程化人工皮肤胶原蛋白含量和合成能力呈上升的趋势。结论利用壳多糖作为基质网架制备的组织工程化人工皮肤具有合成和分泌胶原蛋白的能力。
Objective To analysis the dynamic change of synthesis and content of collagen which is an extra cellular matrix of tissue engineering artificial skin. And the results of collagen analysis might provide a biochemistry basis for the research of tissue engineering artificial skin. Methods Tissue engineering artificial skin was reconstructed by chitosan scaffold, then the synthesis of collagen was detected by the method of ^3H-Pro incorporate and the dynamic state change of collagen content was detected by the method of Woessner. Results The collagen content and collagen synthesis potency of tissue engineering artificial skin showed an increased tendency with the processing of culture. Conclusion The tissue engineering artificial skin which is reconstructed by chitosan as scaffold has the ability of synthesis and secretes collagen.
