视黄酸受体拮抗剂对脐血淋巴细胞免疫球蛋白M生成的作用 被引量:1

Role of Retinoic Acid Receptor Antagonist on Immunoglobulin M Synthesis by Cord Blood Lymphocytes

摘要 目的通过采用视黄酸受体(RARα)选择性拮抗剂Ro41-5253(Ro)进行阻抑实验,以进一步明确视黄酸(RA)对脐血淋巴细胞IgM生成的促进作用是否通过RARα途径调节。方法分离脐血淋巴细胞进行体外培养,在丝裂原SAC活化的同时,加入RARα激活剂RA或(和)受体拮抗剂Ro。检测培养24 h和48 h的细胞RARα,IL-4I、L-6的基因转录水平及培养d5上清中IgM的质量浓度。结果Ro能拮抗视黄酸对脐血淋巴细胞IgM生成的促进作用;同时视黄酸对RARα基因转录水平的上调也被拮抗剂Ro抑制;Ro拮抗视黄酸对两种细胞因子基因转录水平的上调。结论视黄酸对脐血淋巴细胞IgM生成的促进作用是通过RARα受体途径来调控的。 Objective Retinoic acid receptor (RARa) selective antagonist, Ro41 - 5253 (Ro) was used in the counteract test, to confirm the role of RARa in enhancement of IgM synthesis in cord blood lymphocytes (CBLs) by retinoic acid (RA). Methods CBLs were cultured in vitro and stimulated with or without RARa agonist RA and(or) antagonist Ro. The cells were harvested at 24 hours and 48 hours of culture time to measure the levels of gene expression of RARa, IL - 4 and IL - 6. ELISA was used to measure the concentration of IgM in the supernatant of 5 days culture. Results Ro could inhibit the promotion of RA on IgM synthesis in CBLs. RA could up - regulate RARa gene expression, which could be restrained by Ro. Ro also could counteract the up- regulation of RA on IL- 4 and IL- 6 gene expression. Conclusion The effect of RA on IgM synthesis in CBLs is modulated by RARa.
