摘要
目的:探索蔡氏扶正消癥汤抑制胃癌血管生成的机制。方法:用四甲基偶氮唑蓝(MTT)还原法检测蔡氏扶正消癥汤对人胃癌细胞SGC-7901体外增殖的影响;用免疫组化S-P法检测对肿瘤血管内皮细胞生长因子(VEGF)表达的影响。结果:蔡氏扶正消癥汤生药在实验药物浓度范围内,随着剂量的增加,抑制作用更加明显;同一剂量下,72h内药物作用时间越长,抑制作用越明显;蔡氏扶正消癥汤浓度为1.1mg/ml时其平均吸光度为(0.096±0.004),与空白对照组平均吸光度(0.125±0.025)有显著差异(P<0.01),并且VEGF平均吸光度随蔡氏扶正消癥汤浓度增加而降低,表明蔡氏扶正消癥汤能抑制VEGF的表达。结论:蔡氏扶正消癥汤能够抑制人胃癌细胞SGC-7901的体外增殖,抑制VEGF的表达,提示该药有抗血管生成作用。
Objective： To study the mechanism of ＂Caishi Fuzheng Xiaozheng Decoction＂ in inhibiting the angiogenesis of gastric cancer. Methods： MTT colorimetric assay was adopted to study the effect of this Decoction on human stomach adenocarcinoma cell line SGC-7901 hyperplasy in vitro; immunohiatochemistry technique was used to detect the effect of this Decoction on expression of VEGF. Results： This Decoction inhibited the growth of SGC-7901 ceils with increasing dose; with the same concentration, the inhibiting action increased with time; at the concentration of 1.1 mg/ml, its average absorbance was （0.096± 0.004）, with a striking difference with （0.125±0.025） in control group （P〈0.01）; and its average absorbance decreased with the increase of the Decoction＇s concentration, indicating that this Decoction can inhibit the expression of VEGF. Conclusion： ＂Caishi Fuzheng Xiaozheng Decoction＂ can inhibit human stomach adenocarcinoma cell line SGC-7901 hyperplasy in vitro and the expression of VEGF to suppress the angiogenesis of gastric cancer.
