摘要
目的:采用血糖钳方法评价调脂降糖片在2型糖尿病大鼠胰岛素抵抗的增敏作用。方法:采用一次性腹腔注射小剂量链脲佐菌素(30mg/kg体重)配合高糖高脂饲料喂养建立2型糖尿病胰岛素抵抗大鼠模型。用调脂降糖片干预治疗,以二甲双胍作为阳性对照。8周后采用血糖钳技术检测大鼠葡萄糖输注率(GIR),并测定大鼠空腹血糖(FBG)、空腹胰岛素水平(FINS)、血清游离脂肪酸(FFA)、总胆固醇(TC)、甘油三酯(TG),胰岛素敏感指数(ISI)。对大鼠的胰岛素敏感性和脂代谢进行评估。结果:与未治疗的2型糖尿病大鼠比较,调脂降糖片大小剂量治疗后大鼠ISI、GIR明显增加,而TC、TG、FINS、FFA显著下降,其作用与二甲双胍基本相同。结论:调脂降糖片可增加2型糖尿病大鼠胰岛素敏感性,并改善糖脂代谢异常。
Objective: To evaluate the efficacy of "Tiaozhi Jiangtang Tablet" on insulin resistance in NIDDM with glucose clamp technique. Methods: Rats model of insulin resistance with NIDDM was established by intraperitoneal injection of low dose of streptozotoein(STZ) once,combining with high-glucose and high-fat diet. The treatment group(T) was treated with "Tiaozhi Jiangtang Tablet", and the control group (M) was given metformin. Eight weeks after intervention, the glucose infusion rate (GIR) was measured with the glucose clamp technique. At the same time, the level of fasting blood glucose (FBG), fasting insulin (FINS), free fatty acids (FFA), total eholesterol(TC), triglyeride(TG), insulin sensitivity index (INS) were examined. Results: Compared with model group, the level of ISI and GIR were significantly higher in T group, while the levels of FINS, FFA, TC, TG were significantly 1ower. There were not significant differences of all indices between T group and M group. Conclusion:"Tiaozhi Jiangtang Tablet" is effective in increasing insulin sensitivity and improving lipid metabolism in IR rat model.
