摘要
目的比较不同剂量吲达帕胺(Ind)和替米沙坦(Tel)联用对自发性高血压大鼠(SHR)的降压作用及对心功能的影响。方法40只SHR随机分为5组:①空白对照组;②低-低剂量组:Ind 0.06 mg/kg.d-1,Tel 3.57 mg/kg.d-1;③低-高剂量组:Ind 0.06 mg/kg.d-1,Tel 7.14 mg/kg.d-1;④高-低剂量组:Ind 0.12 mg/kg.d-1,Tel 3.57 mg/kg.d-1;⑤高剂量单用组:Tel 7.14 mg/kg.d-1,灌胃给药,连续给药4周,尾袖法测收缩压,Langendorff灌流心脏测量心功能。结果低-低剂量联用组,能有效降低SHR的收缩压,降低左室舒张末期压,减少左室重与体质量之比。结论小剂量联合应用吲达帕胺和替米沙坦对SHR的降压作用及心功能的改善与大剂量单用替米沙坦的疗效一致。
Objective The purpose of this research is to study on the effect of SBP reduction and cardiac function of treatment with different dose combination of Indapamide and Telmisartan on SHR （ spontaneously hypertensive rat）. Methods 40 SHR were randomly divided into 5 groups, ①An untreated group ②Low-low dose combination： Ind 0.06 mg/kg and Tel 3.57 mg/kg per day ③Low-high dose combination： Ind 0.06 mg/kg and Tel 7.14 mg/kg per day ④High-low dose combination： Ind 0.12 mg/kg and Tel 3.57 mg/kg per day ⑤High dose of telmisartan： Tel 7.14 mg/kg per day. All SHR had been treated with drugs by gavage method for successive 4 weeks. The tail arterial SBP of rat was measured once a week during the period of triM. Cardiac function was assessed by isolated langendorff heart perfusion. Results All of 4 treatment groups showed significant decrease in SBP （ P 〈 0.01 ）. LV systolic function was improved in Ind-L/Tel-L group compared with control groups （ P 〈 0.05 ） , and the ratio of heart weight decreased significantly by 21% （P 〈0.05 ） in Ind-L/Tel-L group. Conclusions Combination of low dose of indapamide and telmisartan as well as high dose telmisartan alone decrease the SBP of SHR significantly.
