应用seldi-TOF MS技术分析高低转移肺腺癌细胞株差异表达蛋白 被引量:7

摘要 目的分析体外培养的高低转移肺癌细胞株蛋白质表达差异。方法采用表面增强激光解吸离子化(SELDI)蛋白质芯片技术检测了两种肺癌细胞株(肺腺癌细胞高转移株Anip973和肺腺癌细胞低转移株AGZY)的蛋白质谱。分析不同细胞株之间的蛋白质表达差异。每种细胞用WCX2和IMAC3两种芯片检测。采用PBSⅡC型蛋白质芯片阅读机读取数据,获得的数据采用Ciphergen公司的Proteinchip Software 3.2.1软件分析。结果获得WCX2和IMAC3两种蛋白芯片上Anip973和AGZY细胞株蛋白质谱图谱;在对两种肺癌细胞株的蛋白质谱比较时发现,有14个蛋白质在两种肺癌细胞株中出现明显的变化,其中9个差异蛋白在Anip973细胞中高表达,5个差异蛋白在Anip973细胞中低表达。结论高低转移肺腺癌细胞之间蛋白质谱表达比较时有差异蛋向。 Objective To detect the distinct proteins for different types of lung cancer cell lines. Methods We performed surface enhanced laser desorption ionization (SELDI) mass spectrometry using protein chip technology to compare the differentially expressed proteins in two types of lung cancer cell lines (Anip-973 high transferred lung adenocarcinoma cell line and AGZY low transferred lung adenocarcinoma cell line). To analyze the distinct proteins for different types of lung cancer cell lines. We adopted two different types of protein chip arrays, IMAC - Cu and WAX2, for each types of lung cancer cell lines. Protein chips were examined in PBS H - C protein chip reader(Ciphergen Biosystem Inc) and the protein profiling was analyzed by proteinchip software 3.2.1. Results The results showed that 14 distinct proteins were found in two types of lung cancer cell lines with 9 proteins up - regulated and 5 proteins down - regulated in Anip - 973 when compared the changes of expression in different types of lung cancer cells. Conclusion The results suggest that there are distinct proteins in protein profiling, between lung cancer cells.
