肝内胆管癌细胞系ICC-9810与肝细胞癌细胞系SMMC-7721蛋白质组学比较

Comparative study on proteomics of intrahepatic cholangiocarcinoma cell line ICC-9810 and hepatoeellular carcinoma cell line SMMC-7721

摘要 目的比较肝内胆管癌细胞系ICC-9810、肝细胞癌细胞系SMMC-7721及正常肝细胞系L02的蛋白质表达,筛选胆管细胞癌和肝细胞癌的标志蛋白。方法收集体外培养的肝内胆管癌细胞系ICC-9810、肝细胞癌细胞系SMMC-7721及正常肝细胞系L02细胞,细胞裂解液处理后提取蛋白。应用表面增强激光解吸离子化(SELDI)蛋白质芯片技术同时使用IMAC3和WCX2两种芯片检测肝内胆管癌细胞系ICC-9810、肝细胞癌细胞系SMMC-7721及正常肝细胞系L02的蛋白质谱。用PBSII.C型蛋白质芯片阅读机读取数据,采用Proteinchip Software 3.0.2软件分析数据。结果与L02细胞比较,有12个蛋白质在两种肝癌细胞系中均出现明显的变化,其中4个蛋白高表达,8个蛋白低表达。比较两种肝癌细胞系的蛋白质谱,发现7个蛋自在ICC-810细胞中高表达,10个蛋白低表达;11个蛋白在SMMC-7721细胞中高表达。在L02和ICC-9810细胞中去磷酸化蛋白17175Da表达较多,而在SMMC-7721细胞中磷酸化蛋白17255Da表达较多。结论肝内胆管癌、肝细胞癌与正常肝细胞的差异蛋白表达为寻找肝肿瘤标志物提供了重要线索。17255Da蛋白的磷酸化/去磷酸化可能与肝细胞癌的发生有关。 Objective To compare the expression characteristics of proteins in intrahepatic eholangioeellular carcinoma cell line ICC-9810 and hepatocellular carcinoma cell line SMMC-7721 with normal hepatocellular cell line 1.02 and to screen the distinct proteins for eholangiocellular carcinoma and hepatocellular carcinoma. Methods Intrahepatic eholangiocellular carcinoma cell line ICC-9810,hepato-cellular carcinoma cell line SMMC-7721 and normal hepatocellular cell line L02 cells were cultured and collected. All the cells were lysed by cell lysate solution. Surface enhanced laser desorption/ionization (SELDI) mass spectrometry with ProteinChip IMAC3 and WCX2 was performed to compare the expressed proteins in SMMC-7721 and ICC-9810 with 1.02. Protein profiling was examined by PBSII-C Protein Chip Reader and the proteome database was analyzed by Proteinchip Software 3.0.2. Results Twelve distinct proteins were found in SMMC-7721 and ICC-9810 cell lines in which 4 proteins showed higher expression and 8 proteins lower expression than in L02 cell line. Seven proteins showed higher expression and 10 proreins lower expression in intrahepatic cholangiocellular carcinoma cells, and 7 proteins had the increased expression in hepatoeellular carcinoma cells. The dephosphorylated protein 17 175 Da was expressed higher in ICC-9810 and L92 cells ,but the phosphorylated protein 17 255 Da was expressed higher in SMMC-7721 cells. Conclusion The overexpressed and underexpressed proteins in protein profiling could be candidate biomarkers of intrahepatic cholangiocarcinoma and hepatocellular carcinoma. The protein 17 175 Da phos- phorvlated/dephosphorylated mac be related with the occurrence of hepatoeellular carcinoma.
