摘要
[目的]以幼龄昆明小鼠为研究对象,探讨环境和刺激训练对幼鼠智力和脑结构发育的影响,为改进、优化早产儿和高危新生儿治疗环境提供依据。[方法]将同父异母(母鼠为同胞姐妹)具有相似遗传背景的24只三周龄幼鼠随机分为三组。对照组正常喂养;训练组每日给予运动、平衡及各种感官刺激等规范训练;黑暗组即24 h置于黑暗环境,不给任何训练刺激。一月后应用水迷宫跟踪系统检验各种幼鼠的记忆和判断能力,应用显微图像分析和免疫组化方法检测大脑海马区锥体细胞密度和胆碱乙酰转移酶(ChAT)阳性神经元数量。[结果]处于黑暗环境不给任何训练刺激的幼鼠,记忆和判断能力明显低于经过科学训练的幼鼠和正常对照幼鼠(P<0.05),大脑海马区锥体细胞密度和ChAT 阳性神经元数目均减小(P<0.05)。[结论]幼鼠长期处于封闭环境缺乏必要的刺激训练,可影响其学习和记忆和大脑形态发育。早产儿、高危新生儿住院救治的同时,应重视实施智力开发和大脑发育相关的训练及人文关怀措施。
[Objective] To investigate the ellects of environment amd training on learning and memory and morphological structure of hippocampus in neonatal rats. [Methods] 28 Kunming rats that had similar genes(their father was same, their mothers were sisters)were divided into three teams, (A)control group(n= 8) in which the rats were fed regularly, (B) training team (n=8) in which the rats were regularly exercised ( sporting, balancing and stimulating of sense), (C) dark team (n=8)in which the rats were put in the dark boxes. Morris water maze performance was used to test learning and memory after one month. The numher of pyramidal neurons in CA1 were counted by the microscopical image analysis system and the characteristics of choline acetyltransferase (CHAT) in the hippocampus were observed by immunohistochemistry. [ Results] The level of learning and memory of the rats that in the dark environment and without any training was lower than that of the training team and the control team (P〈0.05). The number of pyramidal neurons in CA1 was less than other teams and the CHAT positive cells were decreased in the dark team (P〈0.05) . [Conclusion] Dark environment and the absence of training and simulating can impaire the development of learning and memory and cause loss of CA1 pyramidal neurons and decrease the numher of the CHAT positive cells in neonatal rats. More attention should be paid to the development of learning and memory as well as the the development of brains of the premature and high risk infants in the NICU
