摘要
目的：探讨预先鞘内给予氯诺昔康对福尔马林致痛大鼠行为学的影响。方法：（1）采用序贯法测定预先鞘内给予氯诺昔康对福尔马林致痛大鼠镇痛作用的ED如值。（2）预先鞘内给予氯诺昔康对福尔马林致痛大鼠行为学的影响：雄性SD大鼠24只，随机分为对照组与实验组（鞘内氯诺昔康组IT组），对照组又分为鞘内生理盐水组（NS组）与肌注氯诺昔康组（IM组）。NS组鞘内注射NS20μl后10min足底注射5％福尔马林100μl，IT组与IM组分别鞘内或肌注氯诺昔康120μg（根据ED50值及95％Cl确定）后10min足底注射5％福尔马林100μl。完成处理后，记录缩腿、舔爪时间。结果：（1）预先鞘内给予氯诺昔康对福尔马林致痛大鼠镇痛作用ED50值为114．6μg，95％Cl为103．0～127．4μg。（2）IT组第二相缩腿、舔爪累计时间显著短于NS组与IM组（P〈0．001）；IM组与NS组第一、第二相缩腿、舔爪累计时间无统计学差异（P〉0．05）。结论：预先鞘内给予小剂量氯诺昔康对福尔马林致痛大鼠有镇痛作用。
Objective： To investigate the effect of preemptive intrathecal lornoxicam on the formalin-induced nociceptive behaviors in rats. Methods： （1） Investigation of the ED50（ median effective dose） value of antinociception of preemptive intrathecal lornoxicam by Dixon＇s up-and-down method in the formalin test. （2） The effect of preemptive intrathecal lornoxicam on the formalin-induced nociceptive behaviors in rats. Twenty-four male SD rats were randomly divided into control group （NS）, intramuscular group （IM） and intrathecal group （IT）. 20μl of NS was intrathecally administered in control group followed by subcutaneous injection of 5% formalin 100μl to the hind plantar 10 min later; 120μg/20 μl （results of ED50 and 95% CI） lornoxicam was administered intramuscularly and intrathecally respectively before formalin injection in IM group and IT group. The flinching and licking time of the injected paw were recorded. Results ： （ 1 ） The ED50 value of antinociception of preemptive intrathecal lornoxicam in the formalin test was 114.6 μg, CI 103.0 - 127.4 μg. （2） The flinching and licking time of phase Ⅱ in IT group were significantly shorter than that in NS and IM groups （ P 〈0. 001 ） ; The flinching and licking time of phase Ⅰ and phase Ⅱ in NS and IM groups were not statistically different （P 〉 0.05 ）. Conclusion： Preemptive intrathecal injection with low dose of lornoxicam produced antinociception in the formalin rats.
