摘要
为探讨补肾宁心方对去势雌兔免疫功能的影响。24只3月龄新西兰雌兔随机分为正常对照组、假手术组、去势组(卵巢切除)、中药组(卵巢切除并灌以补肾宁心中药复方),除正常对照组外均于卵巢切除2周后给予高脂饮食,中药组同时灌胃给药,其余各组均灌服等量的蒸馏水,连续3个月。采用MTT法测定外周血淋巴细胞增殖反应;同时采用半定量逆转录多聚酶链反应(RT-PCR)技术检测动物脾脏IL-2、IL-10 mRNA的表达量。结果显示:雌兔去势后,动物外周血淋巴细胞增殖活性较正常对照组明显降低;补肾宁心方灌药后动物的外周血淋巴细胞增殖活性明显增强,补肾宁心方对动物脾脏的IL-2 mRNA表达水平有明显的上调作用,而对动物脾脏的IL-10mRNA表达水平有明显的下调作用。
The effect of Chinese medicine recipe ＂Bushen-ningxin＂ on immune function of the ovariectomized rabbits was investigated in the present study, in which 24 female New Zealand white rabbits of 3 months old were randomly divided into 4 groups; i.e. normal, sham operated, ovariectomized and treated group by ovariectomization and infusion with ＂Bushen-ningxin ＂ recipe. High-cholesterol diet was given to all the rabbits, except those of the normal group, two weeks after operation. While, the treated group was given additionally with medication and the other 3 groups were given with the same amount of distilled water as controls . The lymphocyte proliferation response of peripheral blood in animals was evaluated 12 weeks later hy MTT assay and the semi quantitative RT-PCR assay was employed to analyse the expression of IL-2 and IL-10 mRNA in spleens. In comparison with the ovariectomized groups, it was found that the ＂Bushen-ningxin＂ recipe could significantly increase the lymphocyte prolifeation response of peripheral blood of the experimental animals, and it had also an up-regulatory effect on the expression of IL-12 mRNA, but it showed a down regulatory effect on the expression of IL-10 mRNA. It suggests that the ＂Bushen-ningxin＂ recipe can promote the immune functions of the ovariectomized rabbits and induce the cytokine profile bias from Th2 type to Thl type.
