人肺腺癌多药抗药细胞系LC—3/CDDP细胞内阿霉素聚集量变化

摘要 多药抗药细胞系LC—3/CDDP为人肺腺癌细胞系LC—3经顺铂选择培养8个月建立的细胞系,对顺铂及其他几种抗肿瘤药物具有抗药性。对细胞内阿霉素聚集量测定发现,该细胞系细胞内阿霉素聚集量较亲代细胞明显减少。二性霉素B、芳香维甲酸乙酯可使细胞内阿霉素聚集量增加,两者联合应用基本可使抗药性逆转。细胞内药物聚集减少可能为LC—3/CDDP细胞系对多种抗肿瘤药物产生交叉抗药性的机制之一。 Multidrug resistant subline,LC-3/CDDP,was established by continuously exposing a human pulmonary adenocarcinoma cell line LC-3 to gradually increasing doses of cisplatin for eight months.This subline showed cross-resistance to adriamycin(ADM), vincristine(VCR),5-fluorouracil(5-FU) etoposide(VP-16),and methotraexate(MTX). This study was to investigate intracellular drug concentration of ADM. The results showed that intracellular ADM concentration of LC-3/CDDP cells was more markedly decreased than that of its parental cells,and aphotericin B and RO·13-6298 could significantly increase intracellular ADM concentration in LC-3/CDDP cells.These results indicated that the decrease of intracellular drug concentration might be one mechanism that the resistant subline showed crossresistance to a variety of anti-cancer drugs.
