胃癌和胃癌前病变中Fhit蛋白表达及意义初探 被引量:1

摘要 目的:研究胃癌和胃癌前病变(肠上皮化生、不典型增生)中Fhit蛋白表达的改变,并对其意义进行初步探讨。方法:用免疫组化SP法检测了30例胃癌和癌旁不典型增生、33例肠上皮化生、14例正常对照胃黏膜中Fhit蛋白的表达。结果:胃癌、不典型增生、肠上皮化生Fhit蛋白表达分别为23.07±10.44、26.39±12.75、37.33±8.94,胃癌、不典型增生Fhit蛋白表达显著低于肠上皮化生(P<0.01),而胃癌和不典型增生之间Fhit蛋白表达的差异无统计学意义;有淋巴结转移与无淋巴结转移、早期与进展期胃癌之间Fhit蛋白表达的差异无统计学意义;高分化、中分化、低分化胃癌Fhit蛋白表达分别为28.77±10.20、24.81±10.83、15.22±4.46,高、中分化与低分化之间的差异有统计学意义(高与低相比P<0.01,中与低相比P<0.05)。结论:Fhit蛋白表达减低可能是部分胃癌发生过程中的早期事件,Fhit蛋白减低与胃癌的恶性程度相关,检测Fhit蛋白可能有助于判断胃癌的恶性程度。 Objective:To investigate whether Fhit protein expression is correlated with clinical parameters,such as lymph node metastasis,infiltration depth and differentiation, and to explore the role of Fhit gene in tumorigenesis of gastric cancer. Methods:Fhit protein expression was evaluated in 30 cases of gastric cancer and their corresponding atypical hyperplasia tissues adjacent to turnouts,33 intestinal metaplasia and 14 normal control samples. Results:The positive unit (PU) of Fhit protein expression in gastric cancer,atypical hyperplasia and intestinal metaplasia was 23.07 ± 10.44,26.39 ± 12.75 and 37.33 ± 8.94, respectively.The expression of Fhit protein in gastric cancer and atypical hyperplasia was lower than that in intestinal metaplasia (P 〈 0.01).No statistical difference was found between gastric cancers with and without lymph node metastasis or between gastric cancers of early stage and advanced stage in Fhit protein expression.The expression of Fhit protein in gastric cancer of high, moderate and low differentiation was 28.77 ± 10.20,24.81 ± 10.83 and 15.22 ± 4.46,respectively.The expression of Fhit protein in high and moderate differentiated gastric cancer was higher than that in low differentiated gastric cancers (for the former P 〈 0.01 ,for the latter P 〈 0.05). Conclusion.Decreased Fhit protein expression may be an early event in the tumorigenesis of some gastric cancer and correlates with malignancy degree of gastric cancer. Detecting Fhit protein expression status may be useful in assessing the malignancy degree of gastric cancer.
