摘要
目的研究电刺激大鼠脚桥核(PPN)对丘脑底核(STN)神经元放电的影响,探索高频刺激STN改善运动不能的机制,为临床高频刺激PPN治疗帕金森氏病提供电生理学依据。方法应用细胞外记录的方法观察不同频率电刺激(强度0.4mA,波宽0.06m s,时程5 s,频率1Hz、10Hz、50Hz、100Hz、130Hz、150Hz、200Hz、250Hz、300Hz)大鼠PPN对STN神经元放电的影响。结果高频刺激(130Hz)PPN可使大多数STN神经元的放电频率降低(P<0.01),该作用具有频率依赖性。结论高频刺激大鼠PPN对STN主要为抑制作用,揭示高频刺激PPN对帕金森病人的运动症状有治疗作用。
Objective The purpose of this study was to research the effect of the electrical stimulation of the pedunculopontine nucleus (PPN) on the neuronal activities of the subthalamic nucleus (STN) in rats. Methods Electrical stimulation (width 0. 06ms, intensity 0. 4mA, duration 5s, frequency 1Hz, 10Hz, 50Hz, 100Hz, 130Hz, 150Hz, 200Hz, 250Hz, 300Hz) was delivered to the PPN. The firing rates of STN neurons were observed by extracellular recording. Results Electric stimulation of the PPN inhibited the firing rates of majority of the STN neurons (P 〈 0. 01 ) . In response to high - frequency stimulation of the PPN, this effect was in a frequency - dependent manner. Conclusions High - frequency stimulation (HFS) of the PPN induced an inhibitory effect on the spontaneous activity of the STN in normal rats. This result suggests that HFS on PPN should be an effective surgical procedure for the treatment of Parkinson' s disease.
