摘要
将30份手术切除的新鲜结直肠癌组织块直接作原代培养48h后,观察培养上清中免疫抑制因子的存在及特性。结果表明:此癌组织培养上清可抑制人外周血单个核细胞(PBMC)产生肿瘤坏死因子(TNFα)并抑制重组人肿瘤坏死因子(rhTNFα)杀伤靶细胞的活性,两种作用均呈剂量依赖性。对抑制rhTNFα杀瘤活性的因子初步纯化的结果证实,其为分子量约40KD的蛋白质。此外,为保证结直肠癌组织培养不污染,本工作对癌组织培养前的无菌消毒方法做了探索,找出最佳处理方案,即“75%酒精-灭滴灵-多种抗菌素”模式。
The present work is on the observation of the prodution and characterization of the immune suppressor factor derived from fresh colorectal carcinoma tissue cultured supernatant.Thirty surgically isolated fresh colorectal carcinoma specimens were investigated.The results shown that the supernatants from the carcinoma tissue culture inhiblted the production of TNF a in LPS induced human peripheral blood mononuclear cells(PBMC) and the killing effect of recombinant human TNF α(rhTNFα) on the target cells WEHI164 in a dose-dependent manner.The active inhibition factor of rhTNF αwas preliminarilly purified,a protein with a molecular weight of about 40 KD was present.Besides,a method of carcinoma tissue asepsis before culture was investigated,a best treatment schedule was worked out,namely,a model of 75% alcohol-metronidazolevarious antibiotics.
