摘要
目的:建立水飞蓟宾葡甲胺在犬体内血药浓度的高效液相色谱检测方法.方法:在弱碱性条件下.血样用叔丁基甲醚进行液-液提取.结果:犬血浆中的成分对水飞蓟宾葡甲胺的检测没有干扰,且水飞蓟宾葡甲胺异构体得到很好分离。线性范围为0.1~5.0μg·mL^-1,高中低3个浓度的平均绝对回收率为(94.2±6.12)%,方法回收率为(104.6±11.05)%。结论:该方法简单准确,灵敏度高,适用于水飞蓟宾葡甲胺体内药动学及临床药理学研究.
AIM: To establish an RP-HPLC method for the determination of silybin meglumine in dog plasma. METHOD: To extract silybin meglumine by methyl t-butyl ether from the plasma in the weak base solution. RESULTS: Silvbin meglumine was separated with no interference found and the two isomers were separated finely. A good linear relationship was obtained between 0.1 - 5.0 μg· mL^-1 ( r = 0.9972). The average absolute recovery of low, medium and high concentration was (94.2 ± 6.12) %. The average method recovery was (104.6 ± 11.05) %. CONCLUSION: This method is simple, sensitive and accurate. It can be widely used in the pharmacokinetie and clinical research of silybin meglumine.
