漏芦水提物对D-半乳糖中毒大鼠急性肝损伤的保护作用 被引量:7

Protective Effects of Rhaponticum uniflorm(L.)DC.Aqerous Extract on Acute Hepatic Injury in Rats Induced by D-galactoasmine

摘要 目的研究漏芦水提物对D-半乳糖所致大鼠急性肝损伤的保护作用。方法50只大鼠随机分为正常组、模型组、大、小剂量给药组、阳性对照组5个组。给药组按10,5 g/kg剂量,阳性对照组按0.2 g/kg联苯双酯灌胃,正常组、模型组以等体积生理盐水灌胃,共5 d。第4天,模型组和给药组按0.5 g/kg剂量腹腔注射D-半乳糖,正常组腹腔注射等体积生理盐水。腹腔注射D-半乳糖24 h后,摘眼球取血,用比色分析法测定大鼠血清天冬氨酸转氨酶(aspartate transam inase,ALT)、丙氨酸转氨酶(alan ine transam inase,AST)的活性,血清、线粒体超氧化歧化酶(superoxide d ismutase,SOD)、谷胱甘肽过氧化物酶(glutath ione perioxidase,GSH-PX)的活性及丙二醛(m alond ialdehyde,MDA)的含量。结果漏芦水提物显著降低因D-半乳糖所致急性肝损伤大鼠血清ALT、AST的升高;对急性肝损伤大鼠血清、线粒体SOD,GSH-PX的活性有明显的升高作用及降低MDA的含量。结论漏芦水提物对D-半乳糖所致大鼠急性肝损伤有保护作用。 Objective To study the protective effects of Rhaponticum uniflorm (L.)DC. aqerous extract on acute hepatic injury in rats induced by D-galactoasmine. Methods 50 rats were randomly divided into normal group, model group, low and high dosage group and Biphengldimethylesteratis group, and the Rhaponticum uniflorm( L. )aqerous extract was perfused to be into stomach for 5days. On the fourth day, the D-galactoasmine 0.5 g/kg was injected into peritoneum in test groups one time to induce the models of acute liver lesion. Colorimetric method measured the activities of aspartate transaminase( AST), alanine transaminase( ALT), superoxide dismutase(SOD) , glutathione perioxidase( GSH-PX), and the content malondialdehyde(MDA) were measured. Resuits Rhaponticum uniflorm( L. ) aqerous extract significantly reduced D-galactoasmine inducing acute hepatic injury by decreasing ALT and AST in serum; the activities of SOD, GSH-PX in test group were higher than those of model group and the content MDA in trcat group were lower than those of model group in serum and liver mitochondia. Conclusion Rhaponticum uniflorm ( L. ) aqerous extract have protecting effects for rats acute hepatic injury by D - galactoasmine.
