高效液相色谱法测定苜蓿中苜蓿素的含量 被引量:7

摘要 目的:建立苜蓿中苜蓿素的 HPLC 测定方法。方法:色谱柱为 Zorbox SB C_(18)(4.6 mm×250 mm,5 μm),以甲醇-水(65:35)为流动相,流速为0.8 mL·min^(-1),检测波长为349 nm,进样量为5μL。结果:苜蓿素进样量在0.02~0.5 μg的范围内与峰面积积分值有良好的线性关系,r=0.9999。高、中、低3个浓度加样平均回收率(n=2)分别为97.8%,97.0%,100%;其偏差分别为0.57,0,90,0.80;平均回收率为98.3%(n=6)。结论:本方法操作简便,结果准确可靠。 Objective:To establish an HPLC method for the determination of tricin in Medicago sativa L. Methods: The HPLC column was C18 (4. 6 mm × 250 mm ,5 μm) ; the mobile phase was methanol - water( 65:35 ) ; the flow rate was 0. 8 mL·min^-1;the detection wavelength was 349 nm;the amount of injection was 5 μL. Results:The calibration curves of tricin were linear in the range of 0. 02 - 0. 5 μg ( r = 0. 9999 ). The recoveries ( n = 2) of three different concentration added in samples were 97. 8%, 97.0%, 100%, respectively; the deviation were 0. 57,0. 90, 0. 80, respectively; the average recoveries ( n = 6) were 98.3 % o Conclusion: The method is rapid, accurate and reliable.
