摘要
目的制备具有肝靶向性的高分子载体乳糖化聚天冬酰肼(Lacto PAHy)并研究其理化性质。方法以氰基硼氢化钠为还原剂,通过还原胺化使乳糖与聚天冬酰肼进行共价结合;利用硫酸苯酚法和聚氨基酸标准曲线法测定乳糖化聚天冬酰肼中乳糖和聚天冬酰肼的含量;利用乌氏黏度计法测定聚天冬酰肼和乳糖化聚天冬酰肼的黏度。结果较佳反应条件为pH 8.9缓冲液介质和24 h的反应时间,此条件可制得乳糖质量百分含量为56%的乳糖化聚天冬酰肼;乳糖化聚天冬酰肼水溶性良好;聚天冬酰肼乳糖化前后的特性黏度分别为0.1621 mL.g-1和0.1847 mL.g-1。结论成功制备了结构和组分确定的乳糖化聚天冬酰肼;硫酸苯酚法和聚氨基酸标准曲线法适合乳糖化聚天冬酰肼的组分分析;乳糖化聚天冬酰肼是一种有望应用于注射给药的高分子载体。
Objective To prepare lactosaminated α, β-polyasparthydrazide(Lacto-PAHy)and investigate its physicochemical property. Methods Lactose was linked with α, β-polyasparthydrazide by reductive amination in presence of sodium cyanoborohydride(NaBH3CN). The contents of lactose and PAHy were measured by the phenol-sulphuric acid method and the standard curve of poly(amino-acid) ; Inherent viscosity was measured. Results The optimal reactive condition included buffer solution of pH8.9 and 24 h reactive time, under which the content of lactose bound was up to 56 % (w) ; Lacto-PAHy was completely soluble in water; Inherent viscositys of Lacto-PAHy and PAHy were 0. 184 7 and 0. 162 1 mL·g^-1 respectively. Conclu- sions The chemical structure of Lacto-PAHy is identified. Phenol-sulphuric acid method and the standard curve of poly(amino-acid)is suitable to analyze the composition of Lacto-PAHy; Lacto-PAHy is a potential polymer carrier for injection.
