摘要
在甘蓝型油菜×花椰菜种间杂种子房离体培养克服远缘杂交不孕的研究中,观察到不同基本培养基、离体培养时杂种子房的发育天数对杂交结实率有明显的影响。结果表明杂交授粉后12d的杂种子房离体培养效果最好,获得15粒种子,其中11粒种子能萌发成苗;培养基试验表明,1/2MS无机+B5有机+IAA1.5mg/L+蔗糖50g/L的培养基效果最好,结籽率达23%,种子萌发率达100%。
Polima cytoplasm male sterlity Brassica napus was as maternal plant to cross with Brassica oleracea var. botrytis. The embryo rescue technique was applied to obtain interspecific hybrids. Ovaries of interspecific hybrid with different days after pollination were cultured in different culture media. It is showed that ovaries taken 12 says after pollination appeared most efficient for seed and plantlet production. Altogether 15 seeds were obtained in experiment and eleven of them germinated on MS medium and grew into plants. Of the medium studied, a half MS medium gave the most satisfactory results for ovary culture and the more plantlet were obtained.
