摘要
目的探讨猪乳一高分子量蛋白质B(HMWP-B)是否来自于血液以及是否为猪乳所特有等问题。方法利用兔抗HMWP-B的多克隆抗体,通过免疫印迹实验来检测泌乳母猪血液中是否存在HMWP-B。结果实验结果证实HMWP-B在泌乳母猪血液中不存在,由此推测HMWP-B可能为来源于乳腺细胞自身合成而非血源性的乳蛋白;同时,结果还表明牛乳和兔乳中不存在HMWP-B,提示HMWP-B可能为猪乳所特有的乳蛋白。结论本实验为证明HMWP-B是由猪乳腺细胞自身合成而不是来自于血液的乳蛋白提供了一定的证据,这为深入开展该蛋白质的功能研究奠定了基础。
[Objective] High molecular weight proteins (HMWP) in sow milk showed four variants of A, B, C, and D by SDS-PAGE, the origin of HMWP-B was studied by this paper. [Method] The experiment is carded out by immunoblotting with the rabbit anti- HMWP-B polyclonal antibody. [Results] The results show that there is no HMWP-B in the serum of the sow, and suggest that HMWP-B is synthesized and secreted by mammary cell. Besides, the result of immunoblotting of cow and rabbit milk with the rabbit anti- HMWP-B polyclonal antibody indicated that there is no homologous protein of HMWP-B in cow and rabbit milk. So, it was presumed that HMWP-B might only exit in sow milk. [Conclusion] The result of this study provides a proof for the hypothesis that HMWP-B is not originated from the blood.
