摘要
运用化学诱变剂MNNG(N-甲基-N′-硝基-N-亚硝基胍)处理坛紫菜(Porphyra haitanensis)原生质体,观察不同剂量的诱变剂对体细胞生长分裂速度和发育分化方向的影响.结果显示;不同剂量的MNNG(7.5~120.0μg·cm^-3)对坛紫菜体细胞的生长都有明显的抑制作用,而且会影响坛紫菜体细胞的发育分化方向.7.5μg·cm^-3、15.0μg·cm-3诱变处理组向畸形细胞团和丝状体发育;30.0μg·cm^-3和60.0μg·cm^-3。诱变处理组向团型叶片发育;120.0μg·cm^-3诱变处理组后期多发育为正常型叶片.同时在体细胞培养过程中发现一种与条斑紫菜(Porphyra yezoensis)单孢子结构功能相似的类单孢子细胞.
Protoplasts of Porphyra haitanensis were treated with chemical mutagen MNNG(N-Methy-N′-Nitro-N-Nitrosoguanidine) to detect the effect of MNNG on the growth and differenciation of the cells. The protoplasts of P. haitanesis prepared by snail digest enzymes were exposed in the concertration from 7.5 to 120.0 μg ·cm^-3f MNNG. The results showed that the LD50 (lethal density 50%) was 14.9 μg · cm^-1 after 6 d exposure. It was observed that at the concentration of 7.5 -120.0μg · cm^-1 MNNG inhibited the growth of Porphyra haitanensis somatic cells distinctively,and the effect of MNNG on the development of P. haitanesis had a dose-effect. At low concentration (7.5 μg · cm^-3 ,and 15 μg · cm^-3) ,MNNG induced the cells developing to the abnormal cell group and protonema. At medium concentration(30 μg · cm^-3 ,and 60 μg ·cm^-1) , MNNG induced the cells developing to the gobbet thallus. It was also detected that in the late period of the culture, the percentage of the normal thallus at the highest concentration( 120 μg· cm^-3) was much than that in other groups. Meanwhile,a special cell type, similar in the structure and function to "monosporelike" cells found in Porphyra yezoensis were observed in the study.
