金丝桃素体外抗巨细胞病毒效应的实验研究 被引量:15

摘要 目的:探讨金丝桃素单体溶液体外抗人巨细胞病毒(HCMV AD169)的效应。方法:采用细胞病变效应(CPE)抑制法观察金丝桃素单体溶液对HCMV AD169毒株的抑制作用,使用MTT比色法检测细胞的损伤程度,并利用吸光度(A)值评价金丝桃素单体溶液的抗HCMV的效应。结果:金丝桃素单体溶液半数中毒浓度(TC50)为195.96 mg/L,半数抑制浓度(IC50)为19.15 mg/L,治疗指数(TI)为10.23。金丝桃素单体溶液具有抗HCMV的作用,且随药物浓度的增高其作用而增强,在6.25-100 mg/L浓度时对巨细胞病毒均有明显抑制作用。结论:体外实验证实,金丝桃素单体是理想的抗HCMV中药。 Objective: To study the inhibitory effect of the solution of monomeric hypericin on human cytomegalovirus(HCMV) in vitro. Methods: Cytopathic effect (CPE) method was used to examine the inhibitory effect of monomeric hypericin on human cytomegalovirus AD169 (HCMV AD169) which infected human embryo lung fibroblasts(HELF) in vitro. Methylthiazoletrazolium (MTT) colorimetric assay was used to test injury degree of cells,and the inhibitory effect of solution of monomeric hypericin on human cytomegalovirus (HCMV) was evaluated by measuring the value of absorbance (A). Results: The median toxic concentration(TC50), median inhibition concentration(IC50 ) and therapeutic index (TI) of the monomeric hypericin solution were 195.96 mg/L, 19.15 mg/L and 10. 23 respectively. It suggested that the solution of monomeric hypericin could inhibit HCMV,and the inhibition effect would be strengthened with the increasing dose of hypericin. The inhibitory effect of the solution of monomeric hypericin at the concentration of 6. 25-100 mg/L was significant. Conclusion: Hypericin is an ideal traditional Chinese medicine on inhibiting human cytomegalovirus in vitro experimentation.
