摘要
对玉米品种“丹2100”和“沈单16”的抗盐性及其分子标记进行了比较研究。生理指标测定结果表明:“沈单16”的超氧化物歧化酶(SOD)活性、过氧化物酶(POD)活性、可溶性糖含量等各指标均高于“丹2100”;低盐胁迫下SOD活性、POD活性升高,高盐胁迫下SOD活性、POD活性下降。盐胁迫情况下,“沈单16”的SOD活性、POD活性均比“丹2100”高,有较强的抗性。以玉米品种“沈单16”和“丹2100”的叶片为材料,采用CTAB-Ⅱ方法提取玉米基因组DNA,应用RAPD分子标记技术对基因组DNA进行多态性扩增。结果从RAPD反应所用的40个随机引物中筛选出11个适宜引物,扩增出63条谱带,其中11条为差异性谱带,其余引物没有扩增出谱带,被淘汰。
The results of the comparative studies between maize varieties Dan2100 and Shendanl6 in their resistance and molecular marker are that: The activities of POD, SOD, and the content of soluble sugar in Shendanl6 are higher than those of Dan2100. The activities of SOD and POD enhance under low salt stress but lower down under high salt stress. The activities of enzyme in Shendanl6 are high. Compared with Dan 2100, Shendanl6 has higher resistance. Leaves of the two cultivars were used as materials. Total DNA was extracted using Cetyl Trimethy Lammonium Bromide(CTAB). Then total DNA was amplified by the Random Amplified Dolymorphism DNA(RAPD)technology. The results showed that: 11 random sequence primers were selected by RAPD technology. 63 bands of DNA were amplified, and 14 different bands were amplified by 6 primers, the others were eliminated because of their failure in generating polymorphism bands.
