摘要
首次采用脱氨再生几丁质凝胶亲和柱层析法直接从鸡蛋清及菜心的茎叶粗提液中纯化了溶菌酶.纯化倍数分别为42.4倍和310.9倍;回收率为70.2%与61.0%;每克酶蛋白的活性分别为99153U和71100U.每克吸附剂对鸡蛋清溶菌酶的活性吸附量为312.7U;蛋白吸附量为16.1mg.两种纯化的溶菌酶经SDS-聚丙烯酰胺凝胶圆盘电泳结果均显示单一条蛋白带.
An efficient and simple chromatographic method was developed for purification of lysozme,utilizing a novel adsorbent-deaminated regenerated chitin. The lysozymes from fresh egg white and crude extract of Brassica campestris L.ssp.chinensis var. utilis stems and leaves were purified 42 4 fold and 310 4 fold,the specific activity reached 99153 U·g -1 and 71100U·g -1 and the recovery of total activity were 70 2% and 61 0% respectively,just by one step. The adsorbing capacity of deaminated regenerated chitin for lysozyme from egg white was 312 7 U·g -1 and 16 1 mg·g -1 . Subjected to SDS polyacacrylamide disc gel electrophoresis both purified lysozymes showed a single protein band.
