摘要
本文对三种不同类型的出血热疫苗的免疫性进行了比较研究,其中沙鼠肾细胞疫苗由β—丙内酯灭活不加任何佐剂,地鼠肾细胞和鼠脑提纯疫苗由福尔马林灭活加氢氧化铝佐剂。三种疫苗经抗原量测定结果只有沙鼠肾细胞疫苗能测出较高的血凝素。家兔经三种疫苗免疫二针后只有沙鼠肾细胞疫苗能测出血抑抗体。中和抗体测定的结果亦以该疫苗的抗体反应最好,全部阳转,滴度也最高(1:20—≥320)。对家兔和长爪沙鼠免疫后用野鼠型病毒攻击结果,三种疫苗均有一定的保护作用,但仍以沙鼠肾细胞疫苗的保护力最强。
The immunogencity was studied for three types of experimental inaetiva ted haemorrhagic fever with renal syndrome (HFRS)vaccine, two tissue cul ture vaccines made from Mongolian Gerbil kidney and hamster kidney and one purified mouse brain vaccine. The M Gerbil kidney cell vaccine was made with Z_(10) virus strain (Apodemus type )and inactivated by β-propiolactone. The mouse brain vaccine made with LR1 virus strain(Apodmus type)and the hamster kidney cell vaccine made with a combination of Apodmus type virus (A3 or A9) and Rattus type virus(L09) were inactivated by formalin and mixed with aluminium hydroxide adjuvants. The virus antigens tested by ELISA or RPHA methods were detected in all the three types of vaccines, howerer except the M. Gerbil kideny cell vaccine, no HA antigen could be detected in the two others. The neutralizing antibody (Nab) was induced with rather high titer (1: 20—1:≥320) in rabbits injected twice with M. Gerbil kedney cell vaccine, but with low titer(1: 10)by the hamster kidney cell vaccine. Although high titers of ELISA(1: 320—1: 1280) and RPHA(1: 256)antigens were detected in the mouse brain vaccine, no Nab could be detected in rabbits after two doses of immunization with it. The vaccine induced higher Nab also afforded stronger protective effect against HFRS virus infection in rabbits and M Gerbils. It seems there is no correlation between contents of ELISA or RPHA antigen in vaccines and the capacity of inducing Nab in animals vaccinated with the vaccine, but it does seem the HA antigen is related somewhat with the immunogeneity. However, the key factors that influence the immunogencity of the HFRS vaccine remain to be further studied
