摘要 目的:采用高效液相色谱法检测发酵液中的替考拉宁含薰。方法:色谱柱为Discovery R C18流动相为乙腈-0.02mol/L乙酸铵,梯度洗脱,柱温30℃,检测波长为277nm。结果:替考拉宁的5个主要成分T-A2-1,T-A2-2,T-A2-3,T—A2—4和T—A2—5在0.2-1.0mg/ml浓度范围内线性关系良好,相关系数(r)分别为0.9996、0.9992、0.9990、0.9994和0.9993。结论:该方法具有测定方便、快捷、结果准确等特点。 Object: To develop a reverse - phase, high - performance liquid chromatography ( RP - HPLC) method for the determination of teicoplanin in fermentation broth. Methods: The separation was performed on an analytical column ( Discovery R Cis, 5μm, 25 cm× 4.6 mm ID ). The mobile phase consisted of acetonitrile - 0.02 mol/L ammonium acetate with gradient elution. The column temperature was kept at 30 ℃ and the detection wavelength was set at 277 nm. Results: There was a good linearity over the range of 0.2 - 1 mg/ml with r of 0.999. Conclusion: The method was convenient, rapid and accurate. It was suitable for online monitoring of teicoplanin during fermentation.
