摘要
采用单层琼脂蚀斑测定法,初步研究了重组痘苗病毒的蚀斑大小与毒力之间的关系。首次发现,在32℃、37℃及40℃下,乙型肝炎-痘苗天坛株和乙型肝炎-痘苗WR株重组病毒在鸡胚细胞上形成的蚀斑,比其相应的未重组病毒为蚀斑明显要小。于小鼠脑内分别接种相同滴度的WR株重组病毒及未重组病毒,两次试验结果LD50分别为10^(-2.36)和10^(-3.76),10^(-2.63)和10^(-4.00),表明重组病毒WR株的小鼠脑内毒力明显低于其未重组株。提示WR株重组病毒的蚀斑大小可能与毒力有关,即蚀斑小毒力较弱,蚀斑大毒力较强。
The relationship between virulence and the size of the plaques of re-combinant vaccinia viruses was studied by plaque assay method on chicken embryo cell monolayer with agar overlay. It was found for the first time that the size of the plaques of rccombinant vaccinia viruses of strain Tian-tan and strain WR is much smaller than those of the parental viruses at the temperature of 32℃, 37℃ and 4℃. WR strain recombinant expressing HBsAg and its wildtype virus were inoculated into the mice brains with preparations adjusted to the same PFU titre. The LD50 of two experiments were 10-2.36 and 10-3.75, 10-2.63 and 10-4.00 respectively. This indicates that the neurovirulence of the WR recombinant is much lower than that of the parental virus and the size cf the plaques of vaccinia virus strain WR may be related to its virulence. Virus with smaller plaques has a weaker virulence than virus with bigger plaques.
