摘要
目的探讨热休克蛋白47(HSP47)在早期糖尿病大鼠的表达及意义。方法雄性Wistar大鼠30 只,随机分为糖尿病模型组(DR组,n=20)及正常对照组(对照组,n=10)。用链尿佐菌素(STZ)腹腔注射制作大鼠糖尿病(DM)模型。于模型建立后1月处死大鼠,取眼球做石蜡切片及视网膜铺片。用胰酶消化视网膜并行 PAS视网膜血管染色,观察管视网膜血管形态变化,用免疫组化法检测HSP47在视网膜的表达。结果正常大鼠视网膜HSP47染色为阴性,但在早期糖尿病大鼠视网膜HSP47染色均为阳性,而视网膜血管形态变化不明显。结论HSP47作为一种胶原特异的分子伴侣,在胶原装配、合成过程中起作用,早期糖尿病大鼠视网膜HSP47表达增加,可能是导致增生性糖尿病视网膜病变(PDR)发展的重要因素之一。
Objective To investigate the expression and significance of HSP47 in the retina of diabetic rats at early stage. Methods Thirty male Wistar rates were randomly divided into the diabetic group ( n = 20) and normal control group( n = 10). Diabetic rats were induced by injection of streptozotocin(STZ) intraperitondally. One month after the model being builded, the eyeballs were removed for making the retinal vascular network, PAS stained, HE stained and immunohistochemically stained. Results The reaction for HSP47 was negative in normal retina. However, HSP47 express in the retina of diabetic rats at early stage. Conclusion HSP47 is a collagen-specific molecular chaperone, the increases of the expression of HSP47 in diabetic retina at early stage indicates that it may be an important cause for the development of proliferative diabetic retinopathy(PDR).
