摘要
采用链脲佐菌素建立大鼠糖尿病 (DM)模型 ,观察 DM形成后 1、2、4周肾小球中纤维连接蛋白(FN)和碱性成纤维细胞生长因子 (b FGF)的表达。结果 :FN在 DM1周和 2周鼠肾小球内的表达无明显变化 ;在第4周 ,FN除了系膜区染色明显加深外 (P<0 .0 5 ) ,还出现了肾小球基底膜的强染。在第 1周 ,模型鼠肾小球系膜区b FGF染色较正常加强 (P<0 .0 5 ) ,第 2周恢复到基础水平 ,第 4周在系膜区又出现了较 1周时更强的表达 (P<0 .0 5 )。上述结果提示 ,DM早期肾小球内即有细胞外基质的堆积 ,FN为一敏感指标 ;b FGF后期表达上调与 FN沉积有关。
We used streptozotocin (STZ) to induce the animal model of diabetes mellitus in rats. After 1 week, 2 weeks and 4 weeks of induction, glomerular expression of fibronectin (FN) and basic fibroblast growth facter (bFGF) were determined by immunohistochemical staining. The results showed that FN staining remained unchanged at 1 week and 2 weeks, but there was a significant increase in the mesangium and glomerular basement membrane at 4 weeks ( P <0.05). A rise in bFGF immunostaining came about in the mesangium of diabetic rats at 1 week ( P <0.05), and it declined to baseline level at 2 weeks, then it increased at 4 weeks to a greater extent as compared with that at 1 week ( P <0.05). This experiment demonstrates that there is extracellular matrix accumulation in early diabetes. FN is a sensitive indicator; the regulation of bFGF is complex, and the increased expression in late stage may play a role in FN deposition.
