摘要
目的研究石菖蒲、赤芍醇提取物对实验性抑郁及脑肠肽的影响。方法通过小鼠悬尾法、游泳绝望法研究受试药物对实验性抑郁行为的影响;建立未预知慢性应激所致大鼠抑郁模型,采用放射免疫方法分别检测血浆、结肠和垂体的血管活性肠肽(VIP)和P物质(SP)含量变化,研究其对中枢性与外周性脑肠肽的影响。结果10min自发活动计数表明,药物不影响动物的自主活动。石菖蒲、赤芍醇提物400mg/kg能显著缩短悬尾和游泳的不动时间;400mg/kg表现出对5-HTP毒性显著增强;200mg/kg、400mg/kg均能显著增强亚致死剂量NA的毒性,导致动物死亡率升高。320mg/kg能显著升高未预知慢性应激所致抑郁大鼠血浆、结肠、垂体VIP含量和结肠SP含量;降低血浆和垂体SP含量,无统计学意义。结论石菖蒲、赤芍醇提物对实验性抑郁具有抑制作用,可对未预知慢性应激所致脑肠肽VIP、SP含量变化产生明显影响,从而提示抗抑郁作用机制与此有关,并为该类药临床治疗抑郁症伴有胃肠道症状提供了依据。
Objective: To explore the effect of Rhizoma Acori Tatarinowii and Radiz Paeoniae Rubra Extract on experimental depression and brain - gut peptide. Methods : Observe the effect on experimental depression by mice tail - hanging test and mice forced swirmning test; setup rats depression model hy unpredictable chronic stimulation, . Determine the content of VIP and SP in plasma, colon and pituitary by the method of radioimmunity, to re,arch the effect on central and periphery brain - gut peptide. Results: The data in 10 minutes spontaneous activity indicates that the ethanolic extract does not disturb the spontaneous activity of the animal.400mg/kg dosage of ethanolic extract can obviously shorten the immobility time in tail - hanging test and lowed swimming test; the 400mg/kg dosage show a eubanced toxicity on 5 - HTP; both dosage of 200mg/kg, 400mg/kg can obviously eubance the toxicity of sub - lethiferous dosage NA, thus induce a higher mortality. The 320mg/kg dosage can obviously increase the VIP content in rats plasma, colon and pituitary and the SP content in colon, under the unpredictable chronic stimulation; Meanwhile, the drug also decease the SP content in plasma and pituitary, which has no meaning in statistics. Conclusions: The ethanolic extract can inhibit the experimental depression and affect the unpredictable chronic stimulation induced content change of VIP and SP; which has provided more choices in clinical therapy on depression go with gastrointestinal syndrome.
