摘要
目的观察大鼠骨髓内皮祖细胞(endothelial progenitor cells,EPC)功能随年龄的改变,探讨年龄因素对EPC功能与活性的影响。方法选择4月龄、18月龄SD大鼠各10例,用密度梯度离心法从骨髓获取单个核细胞,培养7d后激光共聚焦显微镜鉴定DiI标记的乙酰化低密度脂蛋白(DiI-acLDL)和FITC标记荆豆凝血素I(FITC-UEA-I)双染色阳性细胞为正在分化的EPC。采用改良的Boyden小室和粘附能力测定实验观察EPC的迁移和粘附能力。另选取18月龄SD大鼠20只,均分为两组,分别接受4月龄组和18月龄组EPC移植,于移植14d后提取主动脉内皮细胞DNA,检测内皮细胞端粒长度的改变。结果随年龄的增加EPC迁移、粘附及修复损伤内皮功能减退。结论年龄增加可导致大鼠骨髓EPC功能减退。
Objective To investigate the function change of bone marrow-derived endothelial progenitor cells( EPC)in different aging rats, and study the role of age on the function of EPC. Methods Twenty rats were enrolled as donor. Half of them were 4 months old,the other half were 18 months old. Got mononuclearcells from bone marrow. Using chemical analysis after 7 days culture, EPC as measured by DiI-acLDL/FITC-CEA-I double positive cell were detected by laser confocal microscopy. Boyden chamber was used to deter- mine the migration of EPC and the number of recultured EPC was counted to test the adherent function. Another twenty 18 months old SD rats were enrolled as recipient. EPC from different aging donors were transplanted to them. DNA from aorta endothelial cells was extract- ed. Used southern blotting to measure the telomere length. Results The functional ativities of EPC such as migration, adhesion and repair endothelial cells were also impaired in older rats. Conclusion It is suggested that the function of EPC are correlated with ages, the older of age,the worse of EPC functional activity.
