摘要
目的探讨肺癌病人胸腔积液中MAGE-3基因的表达情况,为胸腔积液良恶性的鉴别提供新的诊断线索,进一步为恶性胸腔积液的基因治疗和肿瘤免疫治疗提供研究基础。方法选取20例肺癌合并胸腔积液病人为研究对象,无菌条件下抽取胸水20ml,立即离心取沉淀物,用TRIZOL裂解液抽提RNA,通过RT-PCR方法检测MAGE-3基因的表达情况,以结核性胸水和心衰病人胸水为对照,β-actin为内参照,同步检测基因的表达情况。结果20例肺癌合并胸腔积液病人胸水中检测到MAGE-3基因表达的有14例,阳性率70%,结核性胸水和心衰病人胸水中未检测到MAGE-3基因的表达。结论肺癌病人胸水中MAGE-3基因表达阳性率高,可作为进一步鉴别胸水良恶性的检测指标,为恶性胸腔积液、肿瘤的基因和免疫治疗的研究提供新的线索。
Objective:To investigate MAGE-3 mRNA expression in pleural effusion, and then to give the new evidence of diagnosis, gene therapy and immunotherapy of the malignant pleural effusion. Methods:20 ml of pleural effusion were obtained from 20 eases of lung cancers. Total RNA was extracted by TRIZOL and eDNA was synthesized. Reverse - transcription polymerase chain reaction( RT - PCR) was performed to detect the MAGE - 3 gene expression, using β - actin as the control of the reaction, tuberculosis(TB) pleural effusion and heart failure eases as the control. Results: Of 20 eases of pleural effusion in lung cancers 14 (70%) were MAGE - 3 - positive and MAGE - 3 - negative in TB and heart failure eases. Conclusion: IMAGE - 3 was positive in pleural effusion of lung cancer. This could be an important evidence of the diagnosis, gene therapy and immunotherapy in malignant pleural effusion.
