酸性染料光度法测定烟草中的烟碱

摘要 试验了多种酸性染料与烟碱的反应,发现在pH=4.5的醋酸—醋酸钠缓冲溶液中,酸性染料溴甲酚绿与烟碱能形成离子缔合物,并能被氯仿所苹取,其最大吸收在42Onm处,烟碱含量在0~12μg/ml范围内符合比耳定律,应用此法测定实际样品,灵敏度高、准确度好、操作简便. A method for the determination of nicotine in tobacco was developed based on the sensitive colour reaction of bromocresol green (BCG) - nicotine (NT)system. In HAc - NaAc (pH =4.5)solution, an ionic associate of BCG and NT could be extracted into chloroform,and measured at 420urn. Beer's law was obeyed in the range of 0 to 12μg/ml for nicotine. The proposed method has been used to determine nicotine in tobacco with satifactory results.
