摘要
目的:通过醋酸棉酚(GAA)对经典白血病细胞系HL-60的研究,探索GAA治疗白血病的可能性.方法:用细胞培养、四唑盐比色试验(MTT)、细胞形态学、DNA电泳及NBT还原试验等方法观察不同浓度GAA促进HL-60细胞凋亡及诱导分化的作用.结果:GAA在浓度>6μmol/L时可显著抑制HL-60细胞的增殖,促进其凋亡,并表现出剂量效应;在诱导分化浓度下(1,4μmol/L),GAA可促进HL-60细胞向成熟粒细胞分化(P<0.05).结论:GAA对白血病细胞HL-60的选择性促凋亡作用表明其可能是一种理想的高效低毒抗白血病药物.
Aibl: To investigate the effects of gossypol acetic acid (GAA) on apeptosis and differentiation in classic human actue promylocytic leukemia cell hne HL-60 and study the possibility to treat leukemia with it. METHODS: The cytotoxic activity of GAA on HL-60 cells was detected by means of MTT assay. Apoptosis was identified and analyzed with the help of transmission electron microscopy, cell cycle, and DNA gel electrophoresis, etc. The differentiation of HL-60 cells into more mature granulocytes was detected by nitro blue tetrazolium (NBT) reduction test. RESULTS: More than 6 p.mol/L GAA could suppress the proliferation and induce the apoptosis in HL-60 cells in a dose-dependent manner. NBT reduction test demonstrated that HL-60 cells could also be induced to differentiate into more mature granulocytes by low-dose GAA (1, 4 μmol/L, P 〈 0.05). CONCLUSION: GAA could selectively induce the apoptosis of leukemia HL-60 cells in a smaller dose than that for human peripheral blood mononuclear cells, which suggests that it may be an ideal drug for leukemia therapy.
