摘要
目的:用单细胞凝胶电泳技术(SCGE)研究不同浓度过氧化氢(H2O2)分别作用于HepG2细胞2h后DNA损伤的剂量效应关系,同时观察硫辛酸对H2O2损伤细胞DNA的保护作用.方法:将细胞分为对照组、损伤组和保护组,依次加入不同浓度的H2O2及硫辛酸,应用SCGE检测H2O2引起细胞DNA损伤.结果:随着H2O2浓度的增加,细胞DNA的损伤程度加重,能使尾长、尾部DNA百分含量和尾距显著增加(P<0.05).硫辛酸能使H2O2损伤细胞的尾长、尾部DNA百分含量和尾距均较对照组减少(P<0.05).结论:H2O2对细胞DNA损伤具有剂量效应关系,硫辛酸可以有效地减少22H2O2引起的细胞DNA的断裂损伤.
AIM: To the study dose-effect relationship between the DNA damage of HepG2 cells and the effect of different concentrations of hydrogen peroxide (H2O2 ) for 2 h respectively, and to observe the protective function of lipoic acid from the DNA damage of cells at the same time. METHODS : HepG2 cells were divided into control group, damage group and protective group, then were added with different concentrations of H2O2 and lipoic acid. Two hours later, the single cell gel electrophoresis (SCGE) was used to detect the DNA damage. RESULTS: The DNA damage of cells was more serious with H2O2 concentration increasing. The tail length, the tail DNA content and the tail moment significandy increased ( P 〈 0. 05 ). Lipoic acid could significantly decrease the tail length, the tail DNA content and the tail moment of damaged cells (P 〈0.05). CONCLUSION: H2O2 causes DNA damage of cells in a dose-effect manner, while lipoic acid can reduce effectively this DNA damage caused by H2O2.
