摘要
目的研究钙通道阻滞剂维拉帕米(Verapamil)对人肾癌细胞株GRC-1增殖和凋亡的影响。方法应用四唑盐 (MTT)法和流式细胞仪检测维拉帕米对人肾癌细胞株GRC-1增殖的影响,透射电镜超薄切片观察细胞超微结构检测细胞凋亡。结果MTT比色实验表明维拉帕米50 μmol·L-1、100μmol·L-1对人肾癌细胞株GRC-1增殖的抑制作用明显。流式细胞仪测定细胞增殖周期维拉帕米使大部分细胞停滞于G0/G1期。透射电镜扫描细胞超微结构显示,维拉帕米能促使人肾癌细胞株GRC-1 的细胞凋亡。结论钙通道阻滞剂维拉帕米能抑制人肾癌细胞株GRC-1的增殖,导致细胞凋亡。
Objective To study the effects of Calcium channel blocker on proliferation and apoptosis of human renal carcinoma cell line GRC-1. Methods Human renal carcinoma cell line GRC-1 was in vitro cultured in medium with or without verapamil ( a calcium channel blocker). MTT assay and flow cytometry (FCM) were used to measure the levels of the proliferation of the cultured human renal carcinoma cell line GRC-1 . The cell apoptosis of human renal carcinoma cell line GRC-1 was observed under the electron microscope . Results MTT assay showed that the proliferation of the human renal carcinoma cell line GRC - 1 could be significantly inhibited by 50μmol/L and 100μmol/L verapamil. The study with FCM showed that the most cells of human renal carcinoma cell line GRC-1 was inhibited by verapamil at G0/G1 phase. The observation of ultrastructure of human renal carcinoma cell line GRC-1 showed that verapamil could induce their apoptosis .Condusions Calcium channel blocker( verapumil ) can significantly inhibit the proliferation of human renal carcinoma cell line GRC-1 and induces their apoptosis
