摘要
利用离心、盐析及多步层析法将鼠睾丸醛糖还原酶（ａｌｄｏｓｅｒｅｄｕｃｔａｓｅ，ＡＲ）分离纯化，纯化的酶经ＳＤＳ－ＰＡＧＥ电泳鉴定示一单一蛋白条带，ＡＲ分子量为３９０００±１０００，ＡＲ底物特异性，按１／Ｋｍ大小依次排列为：Ｐ－硝基苯醛，ＤＬ－甘油醛，Ｄ－葡萄糖醛酸，Ｄ－木糖，Ｄ－半乳糖，Ｄ－葡萄糖。
Aldose reductase was purified to a homogeneity from rat testis with repeated centrifugation,salting out, and DEAE-celluose,hydroxylapatite, sephadex-gel column chromatography.The purified enzytne on SDS-PAGE electrophoresis showed a singleprotein band with the molecular weight of 39000±1000 by comparing with standard molecular weight protein marks. The kinetic constants of aldosereductase were determined for several substrates, the Km valuesfor P-Nitrobenzaldehyde ,DL-glyceraldehyde, D-glucaronate, D-xylose,D-galaetose,D-glucose were 0.07, 0. 123,5.6, 19.8, 120, 205mmol/L (rat),respectively.
